I posted a question earlier that was more generically about OS X builds of Basilisk, but it appears maybe the problem is more specific to Big Sur:

Does anyone have serial port forwarding working in Big Sur? Can you say what you had to do?

Hmm... I haven't tried serial port forwarding since upgrading.
What have you attempted so far?

So far, and I'm totally out of ideas:
- chown the /dev/tty.usbserial-FT53JP031 device to my user, and run BII as me. no luck.
- run BII as su: no luck.

Do you have any luck if you instead set seriala to a text file or a bluetooth device?
Re: Basilisk II Serial Ports & Big Sur: Anybody have this working?
I forgot I had tried the bluetooth device as well. Same luck.
I tried pointing at a file on my desktop that I knew my user had perms to, but same luck. No modem or printer port available.

Not sure, just a thought:
Have you tried adding BasiliskII to the list of apps that are allowed to control your computer in System Preferences > Security & Privacy > Accessibility ?

Just tried that now. Also gave it full disk access, which might have been a thing. Results are the same though.
